碧云天生产的冰冻切片快速抗原修复液(Quick Antigen Retrieval Solution for Frozen Sections)是一种特别适合用于冰冻切片的快速抗原修复液,只需室温孵育5分钟即可完成对冰冻切片样品使用多聚甲醛、甲醛或其它醛类试剂固定后的抗原修复。
细胞或组织用多聚甲醛、甲醛或其它醛类试剂固定后,会导致蛋白之间的交联(cross-link),从而遮蔽样品的抗原位点,导致免疫染色时染色信号减弱,甚至出现一些假阳性染色结果。
本抗原修复液含有了广泛使用的SDS等抗原修复试剂等,pH值约为7.4,可以快速有效地去除醛类固定试剂导致的蛋白之间的交联,充分暴露石蜡切片等样品中的抗原表位,从而大大改善免疫染色效果。
通常石蜡切片都需进行抗原修复处理,而冰冻切片可以不进行抗原修复处理。抗原修复会大大改善石蜡切片的免疫染色效果,但对于冰冻切片的染色效果很多文献资料表明也有显著改善。特别是当冰冻切片免疫染色效果欠佳时,可以考虑尝试进行抗原修复。从原理上来看,无论冰冻切片还是细胞爬片等,只要是用多聚甲醛、甲醛或其它醛类试剂固定的样品,进行抗原修复都会有效去除蛋白之间的交联,充分暴露抗原表位,从而大大改善免疫染色效果。
本产品特别适合用于冰冻切片的抗原修复并用于后续的免疫染色。

一个包装的本产品可以配制成500毫升抗原修复液(1X)。按照每个片子需要10毫升抗原修复液(1X)计算,一个包装的本产品可以用于50个样品。按照每个片子需要5毫升抗原修复液(1X)计算,则一个包装的本产品可以用于100个样品。

注意事项:
抗原修复过程可以使用碧云天的染色缸和染色架或邮寄夹进行操作。塑料染色缸、染色架和邮寄夹可以很好地耐受沸水浴,而玻璃染色缸需避免骤冷骤热导致的玻璃破碎。
本抗原修复液使用前必须先充分溶解并混匀,然后用重蒸水或Milli-Q水稀释5倍,配制成抗原修复液(1X)。
本产品仅限于专业人员的科学研究用,不得用于临床诊断或治疗,不得用于食品或药品,不得存放于普通住宅内。

使用说明:
1. 对于石蜡切片:
a. 脱蜡:切片在二甲苯中脱蜡5分钟,再换用新鲜的二甲苯脱蜡,共用二甲苯脱蜡3次。无水乙醇5分钟,两次。90%乙醇5分钟,两次,70%乙醇5分钟,一次。蒸馏水5分钟,两次。
b. 抗原修复:用重蒸水或Milli-Q水将本抗原修复液(5X)稀释5倍,配制成抗原修复液(1X),例如1ml本抗原修复液(5X)加入4ml重蒸水或Milli-Q水,混合均匀,即得5ml抗原修复液(1X)。将切片浸泡在抗原修复液(1X)中,室温孵育5分钟。用免疫染色洗涤液洗涤3-5次,每次3-5分钟,以充分去除残留的SDS等试剂。随后即可进行封闭等后续的免疫染色步骤。
2. 对于冰冻切片:
用免疫染色洗涤液洗涤切片5分钟。将切片浸泡在抗原修复液(1X)中,室温孵育约5分钟。用免疫染色洗涤液洗涤3-5次,每次3-5分钟,以充分去除残留的SDS等试剂。随后即可进行封闭等后续的免疫染色步骤。
3. 对于其它样品的抗原修复,可以参考石蜡切片或冰冻切片的步骤进行。
